Xxxii. the Preparation and Use of the Bone Phosphatase. by Marjorie Martland And

چکیده
THE phosphatase obtained from the bones of growing animals has proved very useful in the study of the chemical composition of phosphoric esters of biochemical interest. By its aid the phosphoric acid groups may be quantitatively removed under conditions of pH and temperature which do not involve the rupture of other linkages in the molecule such as would occur during acid hydrolysis. For such purposes, as well as for the study of the mode of action of the enzyme, it is desirable to have stable preparations of the phosphatase, of high activity and as free as possible from blood pigments and such other impurities as would be difficult to remove from the hydrolysis products. The following is a brief account of experiments carried out with this object in view, and of the methods at present employed.

Quantification and comparison of bone-specific alkaline phosphatase with two methods in normal and paget’s specimens
Background: Bone-specific alkaline phosphatase (BAP) is synthesized by the osteoblasts and is presumed to be involved in the calcification of bone matrix, though its precise role in the formation process is unknown. The aim of the present study was to measure the BAP activity in Paget's and normal specimens by two different techniques. Methods: Total ALP (TAP) as well as BAP activity-measuring ...
متن کاملXci. the Possible Significance of Hexose- Phosphoric Esters in Ossification
IN the earlier papers of this series [Robison and Soames, 1924] some account was given of the properties of the phosphatase which is present in ossifying cartilage, bones and teeth, and also in the kidney and intestinal mucosa. In the following we give the results of a more detailed study of some of its properties about which further knowledge was desirable in order to understand the part playe...
متن کاملP-178: Separation and Identification of Alkaline Phosphatase Isozymes during Pregnancy
Background: Alkaline phosphatase (ALP), (EC 3.1.3.1) is a hydrolase enzyme responsible for removing phosphate groups from various molecules in the body. In humans, ALPis present in all tissues such as liver, bile duct, kidney, bone, and the placenta which detection of its activity is so useful in molecular biology. Pregnancy is associated with normal physiological changes that assist fetal surv...
متن کاملPreparation and investigation of polylactic acid, calcium carbonate and polyvinylalcohol nanofibrous scaffolds for osteogenic differentiation of mesenchymal stem cells
Objective(s): In this study, the effect of electrospun fiber orientation on proliferation and differentiation of mesenchymal stem cells (MSCs) was evaluated. Materials and Methods: Aligned and random nanocomposite nanofibrous scaffolds were electrospun from polylactic acid (PLA), poly (vinyl alcohol) (PVA) and calcium carbonate nanoparticles (nCaP). The surface morphology of prepared nanofibrou...
متن کاملImpact of Ionizing Radiation on the Expression of CDC25A Phosphatase (in vivo)
Background and Objective: The cell division cycle 25 (CDC25)is a familyof highly conserved dual-specificity phosphatases that activate cyclin-dependent kinase complexes. These complexes are the main cell cycle regulators. Mammalian cells ,exposure to DNA damaging radiations such as ionizing radiation and ultraviolet light, prevent cell cycle progression by activation of checkpoint pathways an...
